چکیده

Eg5, the product of Kif11 gene, also known as kinesin spindle protein, is a motor protein involved in proper establishment bipolar mitotic spindle. Eg5 one 45 different kinesins coded human genome superfamily. Over last three decades has attracted great interest promising new target. The identification monastrol specific inhibitor ATPase activity domain inhibiting microtubule motility vitro and cellulo sparked an intense academia industry to pursue novel small molecules that target order be used cancer chemotherapy based on anti-mitotic strategy. Several inhibitors entered clinical trials. Currently field faced with problem most tested exhibited only limited efficacy. However, inhibitor, Arry-520 (clinical name filanesib), demonstrated efficacy patients multiple myeloma scheduled enter phase III At same time, trends research are emerging, including increased binding sites focus drug synergy established antitumor agents improve chemotherapeutic This review presents updated view structure function Eg5-inhibitor complexes, traces possible development resistance their potential therapeutic applications, surveys current challenges future directions this active discovery.

Targeting mitotic kinases for anti - cancer treatment

Cancer cells proliferate unlimitedly. Therefore, targeting the proliferative capacity of cancer is one of the smartest ways to treat cancer. In this vein, understanding the basis of mitosis is essential for the advanced treatment of cancer, let alone its importance in basic biology. Indeed, recent progress in anti-cancer strategy has come from the development of a group of inhibitors targeting ...

Eg5 inhibitor YL001 induces mitotic arrest and inhibits tumor proliferation

Eg5 is a kinesin spindle protein that controls chromosomal segregation in mitosis and is thus a critical drug target for cancer therapy. We report the discovery of a potent, selective inhibitor of Eg5 designated YL001. YL001 was obtained through shape similarity based virtual screening, and it bears a 1,5-disubstituted tetrazole scaffold. YL001 exhibits favorable bioactivity in a variety of can...

New Anti-Influenza Agents: Targeting the Virus Entry and Genome Transcription

Introduction: The emergence and spread of the pandemic H1N1 influenza virus in 2009 indicates a limitation in the strategy to control the infection, despite a long-established vaccination programme and approved antivirals. Production the proper vaccine against influenza is difficult due to the genetic recombination of virus in the event of pandemic and co-circulation of drug-resistance variants...

Interaction of the mitotic inhibitor monastrol with human kinesin Eg5.

The microtubule-dependent kinesin-like protein Eg5 from Homo sapiens is involved in the assembly of the mitotic spindle. It shows a three-domain structure with an N-terminal motor domain, a central coiled coil, and a C-terminal tail domain. In vivo HsEg5 is reversibly inhibited by monastrol, a small cell-permeable molecule that causes cells to be arrested in mitosis. Both monomeric and dimeric ...

Targeting Metabolic Symbiosis to Overcome Resistance to Anti-angiogenic Therapy

Despite the approval of several anti-angiogenic therapies, clinical results remain unsatisfactory, and transient benefits are followed by rapid tumor recurrence. Here, we demonstrate potent anti-angiogenic efficacy of the multi-kinase inhibitors nintedanib and sunitinib in a mouse model of breast cancer. However, after an initial regression, tumors resume growth in the absence of active tumor a...
